Sleep and Genetics: Why Poor Sleep Hits Some Brains Harder

Poor sleep may be more harmful to the brains of people who carry certain variants of a gene involved in the brain's overnight waste-removal system, according to new research from Edith Cowan University (ECU) in Australia. The findings, published in Alzheimer's & Dementia, suggest that the interplay between genetics and sleep habits could explain why some people show faster brain decline than others, even with similar overall risk.

The Research

Researchers at ECU's Centre for Precision Health studied 13 common variants of the AQP4 gene, which helps regulate the flow of fluid that supports the brain's natural cleaning process — a process that becomes especially active during sleep. They combined genetic data with self-reported sleep patterns, brain scans, and cognitive tests from a cohort of participants over time (specific sample size not provided in the press release).

The results revealed striking differences: some participants who reported shorter sleep showed faster loss of grey matter — a key indicator of brain health — but only if they carried certain AQP4 variants. In others, taking longer to fall asleep was linked to lower brain volume. Cognitive performance also changed differently depending on the genetic variant and sleep disturbance reported.

Dr. Ayeisha Milligan Armstrong, a researcher involved in the study, emphasized the interaction: "It's not just which genes you carry — it's how those genes interact with the world around you. The same variant can look protective or detrimental depending on how someone is sleeping."

Why It Matters

For years, scientists have known that poor sleep and Alzheimer's risk are linked, but this research suggests the pathway isn't the same for everyone. Dr. Tenielle Porter, another researcher on the team, noted: "Instead of assuming everyone at risk follows the same pathway, a more targeted and personalized approach to Alzheimer's prevention may be needed."

This could eventually lead to personalized recommendations: for someone with a high-risk AQP4 variant, improving sleep might be especially critical, while someone else with a different variant might benefit more from other interventions. However, the researchers caution that their findings need replication in larger, more diverse groups before genetic testing is recommended clinically.

While you can't change your genes, you can improve your sleep habits. Aim for 7–9 hours of quality sleep per night, maintain a consistent sleep schedule, and create a restful environment — dark, cool, and quiet. If you have trouble falling asleep, practice relaxation techniques like deep breathing or meditation. These steps support your brain's overnight cleaning system, which may be particularly beneficial if you carry a higher-risk AQP4 variant.
