Protein SORLA protects brain from Alzheimer's tau damage

A natural protein in the brain may help shield against the toxic tau tangles that drive Alzheimer’s disease and other dementias. Researchers at Sanford Burnham Prebys have discovered that higher levels of the protein SORLA reduce tau accumulation, brain atrophy, and synaptic damage in mice.

The research

Scientists led by Timothy Huang, PhD, at Sanford Burnham Prebys published results on July 17, 2026, in Science Advances. They studied mice genetically modified to develop tau tangles (tauopathies) and crossbred them with mice that produce elevated human SORLA. Mice with extra SORLA showed less tau hyperphosphorylation, fewer tau “seeds” that recruit more protein into clumps, less brain atrophy, and healthier synaptic connections. In contrast, mice lacking the Sorl1 gene (which makes SORLA) had more severe tau damage.

“When you upregulate SORLA, you can suppress the negative effects found in tauopathies,” said lead author Huijie Huang, PhD. The team used advanced sequencing to show SORLA’s effects across both neurons and supporting glial cells.

Why it matters

Alzheimer’s affects over 6 million Americans, and tau tangles correlate closely with cognitive decline. SORLA was already known to reduce amyloid-beta plaques, another hallmark of Alzheimer’s. Now it appears to combat tau damage as well. This dual action makes SORLA a promising target for drugs that could boost its levels or activity, potentially slowing or preventing dementia.
