In a mouse model of Alzheimer's disease, scientists restored more than two hours of lost sleep per day by temporarily removing overactive immune cells—even though the hallmark amyloid plaques remained untouched.
The Research
A team at the University of Kentucky, led by Shannon L. Macauley, Ph.D., and first author Nicholas J. Constantino, Ph.D., published in Alzheimer's & Dementia that microglia—the brain's immune cells—are the main drivers of sleep disruption in Alzheimer's. Using EEG and EMG recordings in mice with amyloid plaques, they found that microglia triggered inflammation that kept the brain from getting deep, restorative sleep. When they gave the mice a drug called Pexidartinib (PLX3397) for 14 days, it eliminated about 87% of microglia. In their absence, the mice regained more than two hours of sleep per day, even though the plaques remained unchanged. Light sheet microscopy confirmed the widespread immune activation. The study included both plaque-forming mice and normal aging controls, examined at six and 18 months.
