Brain organoids grown from Alzheimer’s patients’ own cells revealed striking differences in how individual tissues respond to an SSRI antidepressant. The findings, led by Vasiliki Machairaki at Johns Hopkins Medicine, suggest these pea-sized clusters of brain tissue could one day guide personalized Alzheimer’s care.
What the researchers did
The team reprogrammed blood cells from people with Alzheimer’s disease and from healthy controls into stem cells, then coaxed them to form hindbrain organoids that produce serotonin. The study included hundreds of organoids from multiple patients. They then treated the organoids with escitalopram oxalate, a common SSRI.
What they found
Some patient-derived organoids showed increased proteins involved in serotonin signaling and neuron communication after treatment, while others showed little or no change. Organoids from healthy individuals responded differently. The work, published in Alzheimer’s & Dementia, also identified particles released by organoids that might serve as biomarkers for diagnosing Alzheimer’s or tracking its progression.
