A newly mapped neural circuit shows how eating palatable food releases dopamine in the prefrontal cortex, which then activates a relay of inhibitory neurons to shut down the hyperactive stress cells in the hypothalamus that drive chronic anxiety.
The Research
In a study published in Advanced Science, Dr. TU Jie and colleagues at the Shenzhen Institute of Advanced Technology (SIAT) of the Chinese Academy of Sciences set out to find the physical wiring that connects the brain's reward system to its stress-response machinery. Using mice, they combined conventional behavioral tests with high-resolution 3D behavioral mapping to track movement patterns with extreme precision.
They found that chronic stress triggers severe hyperactivation of corticotropin-releasing factor (CRF) neurons in the paraventricular nucleus (PVN) of the hypothalamus, a core stress center. This overactivity produced measurable anxiety-like behaviors. When the mice consumed palatable food, however, both the neural hyperactivity and the anxious behaviors reversed.
Real-time in vivo recordings revealed the sequence: eating rewarding food causes dopamine release in the prefrontal cortex (PFC), which activates excitatory D1 receptor (D1R) neurons. These PFC D1R neurons then suppress the overactive PVN CRF neurons. Because the cortical neurons are excitatory, they needed an intermediate inhibitory relay. The team identified CRFR1-expressing neurons in the peri-PVN region as that critical relay node. The full pathway—PFC D1R → peri-PVN CRFR1 → PVN CRF—provides a top-down brake that converts a dopamine hit from food into a physical cellular shutdown of chronic stress.
