How Adolescent Brain Remodeling Temporarily Hides Early Memories

Researchers have discovered that a protective structure around neurons in a key memory region temporarily breaks down during late adolescence, making memories formed earlier in life briefly inaccessible before they return in adulthood with less detail.

The research

Scientists at Albert Einstein College of Medicine studied mice to understand how memory circuits mature during adolescence. They focused on the retrosplenial cortex (RSP), a brain region important for contextual memory — remembering the where and when of events.

Published in PLOS Biology, the study found that perineuronal nets (PNNs) — dense, lattice-like structures that wrap around certain neurons and stabilize connections — undergo a dramatic remodeling in the RSP during late adolescence. These nets shrunk and diminished in density, driven by a temporary drop in the growth factor TGFβ2, before rebuilding in adulthood.

Critically, this change was exclusive to the RSP. The hippocampus, a nearby memory center, showed no such PNN fluctuations, indicating this is a targeted maturation process, not general brain decline.

Behaviorally, mice trained in early adolescence to associate a specific environment with a mild fear response struggled to recall that memory when tested weeks later during late adolescence. However, when the researchers artificially reinforced the PNNs or restored normal TGFβ2 levels, the mice immediately regained access to those early memories. The memories were never erased — just temporarily blocked by circuit instability.

When the memories spontaneously resurfaced in adulthood, they had lost their original precision. Instead of remembering the exact context, the adult mice generalized their fear response to new environments. This mirrors the human “reminiscence bump,” where adults vividly recall the emotional essence of youth events but lose the fine details.

Why it matters

This study offers a biological explanation for why memories from early adolescence can feel fuzzy or hard to retrieve during the teenage years, then seem to return later with a different flavor. It also highlights that adolescence is not just about hormones or behavior — it is a period of active, targeted brain remodeling.

The findings align with the developmental window when psychiatric conditions like schizophrenia and major depressive disorder often emerge. The authors suggest that disruptions in this PNN remodeling process could contribute to vulnerability in genetically predisposed individuals. Understanding this mechanism may one day inform interventions, but for now, it underscores that memory access is dynamic and biologically tuned.
