3D Brain Tissue Model Replicates Alzheimer's Pathology

A new three-dimensional human brain tissue model that faithfully reproduces Alzheimer's disease pathology has been developed, opening the door to rapid, automated drug testing. This breakthrough, achieved by researchers at LMU Munich, could accelerate the search for effective treatments.

The Research

Led by Professor Dominik Paquet at the Institute for Stroke and Dementia Research, the team spent nine years perfecting a method to grow miniature brain tissues, or spheroids, from human stem cells. These spheroids, roughly half a pinhead in size, contain three key cell types: neurons that form functional synaptic networks, astrocytes that provide metabolic support, and microglial cells that act as the brain's immune system, actively surveying and clearing debris. Published in Nature Neuroscience, the study confirms that these tissues express genes and proteins relevant to Alzheimer's pathology. To validate the model, the researchers induced amyloid-beta aggregates—hallmark plaques of Alzheimer's—and then administered newly approved therapeutic antibodies. The microglia successfully cleared these plaques, demonstrating the model's clinical relevance. The team is now adapting the platform for robotic manufacturing to produce thousands of identical spheroids for industrial drug screening.
