The Research
Scientists at King's College London used precision functional genomics to study ZNF804A, the first schizophrenia risk gene identified from human genomic data. The study, published in Science Advances (May 2026), pinpointed that ZNF804A is most active during the second trimester of brain development, and specifically in glutamatergic neurons, which are key excitatory cells.
The team used CRISPR-Cas9 gene editing to suppress ZNF804A in developing cortical neurons. This led to two major effects: neurons transported excess ribosomes to the tips of their dendrites, causing local protein translation to accelerate, and this surge increased the density of synaptic signaling proteins, making the synapses far more electrically excitable when chemically stimulated.
Co-senior author Professor Deepak Srivastava explained, 'Large-scale genetic studies identify risk factors, but they don't tell you when the gene is active or in which cell type. Precision functional genomics fills that gap.'
