SC-TauPath: AI Maps Tau Spread in Alzheimer's via Brain Connections

Alzheimer's disease spreads through the brain like a slow wildfire, and a new AI framework can now trace its path using brain scans alone. Researchers developed SC-TauPath, a model that links structural connections to tau protein spread, potentially enabling earlier and more precise tracking of the disease.

The Research

Scientists from the University of Texas at Arlington and collaborators analyzed brain scans from 234 participants in the Alzheimer's Disease Neuroimaging Initiative (ADNI). They combined diffusion tensor imaging (DTI), which maps white matter tracts, with Flortaucipir PET scans, which reveal tau protein deposits. They then trained a neural network augmented with a Network Diffusion Model to predict tau levels in different brain regions based on structural connectivity. By attributing each connection's contribution to tau prediction, they created multi-scale pathway maps—backbone edges, high-traffic routes, and hub regions. Their maps aligned with the well-known Braak staging, which describes tau progression from the entorhinal cortex outward. The study achieved strong cross-validated predictions, demonstrating that structural connections carry specific information about regional tau distribution. The paper, posted on arXiv in June 2026, is authored by Jing Zhang and colleagues.

Why It Matters

Understanding how tau spreads is key to diagnosing and treating Alzheimer's. Current models are either overly complex or lack interpretability. SC-TauPath offers a clear, data-driven way to visualize disease progression from non-invasive scans. It also opens avenues for testing therapies that target specific pathways.
