Blocking a Little-Known Protein Could Slow Alzheimer's, Mouse Study Suggests

Researchers at ETH Zurich have identified a protein that fuels Alzheimer's – and they've developed a compound that blocks its harmful effects in mice, potentially opening a new avenue for treating dementia.

The Research

Ursula Quitterer, Professor of Molecular Pharmacology at ETH Zurich, and her team studied an enzyme called GRK2, which helps cells respond to stress. Using brain tissue samples from dementia patients and non-dementia controls, they found unusually high levels of an inactive form of GRK2 in the brains of people with dementia. In a mouse model of Alzheimer's, they saw the same pattern – and discovered that inactive GRK2 clumps together on mitochondria, the cell's powerhouses, blocking their pores and reducing energy production.

This GRK2 clumping also increased production of amyloid beta, a protein fragment strongly linked to Alzheimer's. The buildup of amyloid beta then triggers more stress, which creates more inactive GRK2 and more clumps – a vicious cycle that drives dementia progression.

To break this cycle, the team synthesized several compounds and tested them in cell cultures and older mice. One compound, named 'Compound 10,' stood out: it prevented GRK2 from clumping, allowing mitochondria to function normally. In treated mice, nerve cell death slowed, amyloid beta levels dropped, and the animals lived longer. Even more striking, the compound improved heart function and reduced signs of aging – older mice even developed fewer grey hairs.

Why It Matters

Alzheimer's is a complex disease, and current treatments target amyloid beta directly, with mixed results. This research suggests that GRK2 might be an upstream trigger – by stopping its clumping, you might prevent the cascade that leads to dementia. The fact that heart health also improved hints that GRK2 could play a role in other age-related conditions.
